If you are saying perek shira and Nishmas as a segula for 40 consecutive days, do u have to say it before shikia? Also, can you say it on shobbos?

Yes, the general definition of day is until sunset (shikiya), so you should do so everyday before that. There is no problem saying perek shira or nishmat on Shabbat.
